Audrain County is located in the central part of Missouri and is part of the Columbia, MO Metropolitan Statistical Area. The county seat is Mexico. The County was organized on January 29, 1836.

The county government is composed of three elected commissioners, a county clerk, a sheriff, a prosecuting attorney, a public administrator, a collector, a treasurer, and a surveyor.

The county is divided into nine townships: Audrain, Benton, Central, Laddonia, Liberty, Martinsburg, Prairie, Rush Hill, and Vandalia.

The county is served by two school districts: Mexico Public Schools and Community R-VI School District.

The county is home to several state parks, including Mark Twain State Park, Cuivre River State Park, and Graham Cave State Park.
