Knox County is located in the northeastern corner of Missouri and is part of the Hannibal, MO-IL Metropolitan Statistical Area. The county seat is Edina. The county was organized on February 14, 1845.

The county is governed by a three-member county commission, elected from single-member districts. Other elected officials include a sheriff, prosecuting attorney, assessor, collector, and public administrator.

The county is part of the Lewis and Clark National Historic Trail and the Santa Fe National Historic Trail. It is also home to the Mark Twain National Forest and the Mark Twain Lake State Park.
